A New Process for Extracting Alginates from Laminaria digitata: Reactive Extrusion ArchiMer
Vauchel, Peggy; Kaas, Raymond; Arhaliass, Abdellah; Baron, Regis; Legrand, Julien.
Alginates are natural polysaccharides that are extracted from brown seaweeds and widely used for their rheological properties. The main step in the extraction protocol used in the alginate industry is that of alkaline extraction. A batch process is used for this step, which is time-, water-, and reactant-consuming. The possibility of extracting by reactive extrusion was investigated. The reactive extrusion process appeared to be more efficient than the batch process for the alkaline extraction of alginates from Laminaria digitata in several key ways: Time demand is reduced from about an hour to only few minutes, water and reactant requirements are divided by more than a factor 2, extraction yield is 15% higher (relative enhancement), and the rheological...

A Physiologically Structured Equation to Consider Quota Heterogeneity in the Droop Model ArchiMer
Mairet, Francis; Baron, Regis.
The Droop model allows to represent microalgae growth limited by a nutrient, using a cell quota (also referred to as variable-yield) approach. Single-cell measurements have revealed quota heterogeneity in phytoplankton collected from field studies. Such heterogeneity can be due, among other factors, to spatial structure (e.g. in biogeochemical cycles in the ocean, or for photobioreactors connected in series). Nonetheless, quota heterogeneity is generally omitted in modelling studies, using an average quota approach, or included in size-structured or individual-based models. Here, we propose a distributed Droop equation to tackle this problem, considering subpopulation growth -in line with Droop macroscopic view- rather than cell division dynamics. We...

Bacterial community structure of the marine diatom Haslea ostrearia ArchiMer
Lepinay, Alexandra; Capiaux, Hervé; Turpin, Vincent; Mondeguer, Florence; Baron, Regis; Lebeau, Thierry.
The marine diatom Haslea ostrearia produces a water-soluble blue-pigment named marennine of economic interest (e.g. in aquaculture for the greening of oysters). Up to date the studies devoted to ecological conditions under which this microalga develops never took into account the bacterial-H. ostrearia relationships. In this study the bacterial community was analysed by PCR-TTGE before and after H. ostrearia isolation cells recovered from 4 localities, to distinguish the relative part of the biotope and the biocenose and eventually to describe the temporal dynamic of the structure of the bacterial community. The bacterial structure of the phycosphere differed strongly from that of the bulk sediment. The similarity between bacteria recovered from the...

Comparative Value of a Sorting Procedure and Quantitative Descriptive Analysis to Investigate the Influence of Processing Parameters: Case Study of Hydrolysate Production From Salmon By-Products ArchiMer
Cardinal, Mireille; Baron, Regis; Kouakou, Christelle; Prost, Carole; Courcoux, Philippe.
Many papers have recently discussed the value of a free sorting method as a rapid and simple alternative to quantitative descriptive analysis, considered the reference tool for food sensorial characterization. The aim of the present paper is to evaluate whether this method of free sorting can also be used to investigate the influence of processing parameters. An experimental design was applied to production conditions of enzymatic hydrolysates from salmon by-products. The effect of four processing parameters (time and temperature of hydrolysis, sugar and antioxidant addition) on the odor of the hydrolysates was studied using a sorting task with 45 untrained panelists and a quantitative descriptive analysis carried out with 11 trained panelists. This study...

Continuous fluorescence recording as a way to improve Pacific oyster (Crassostrea gigas) models of paralytic shellfish toxin accumulation ArchiMer
Baron, Regis; Couedel, Marielle; Joret, Camille; Garen, Pierre; Truquet, Philippe; Masselin, Pierre; Bardouil, Michele; Lassus, Patrick.
A simple system was used to simulate the effect of alternating toxic (paralytic shellfish poisoning toxins) and non-toxic microalgal diets on oyster feeding behaviors and rates of toxin accumulation. These experimental conditions were meant to reflect, to some extent, the incoming and outgoing fluxes of toxic algae observed at the mouth of the Penze estuary (Northern Brittany, France). Physiological and toxicological parameters were estimated based oil fluorescence measurements recorded continuously at the outlet of each experimental tank, which contained a single oyster. Q(tox), this variable describes toxin uptake in oysters, it was used (instead of the toxin ingestion rate): i) in simple graphical analyses, ii) as well as in one- and two-compartment...

Decrease in dynamic viscosity and average molecular weight of alginate from Laminaria digitata during alkaline extraction ArchiMer
Vauchel, Peggy; Arhaliass, Abdellah; Legrand, Jack; Kaas, Raymond; Baron, Regis.
Alginates are natural polysaccharides that are extracted from brown seaweeds and widely used for their rheological properties. The central step in the extraction protocol used in the alginate industry is the alkaline extraction, which requires several hours. In this study, a significant decrease in alginate dynamic viscosity was observed after 2 h of alkaline treatment. Intrinsic viscosity and average molecular weight of alginates from alkaline extractions 1-4 h in duration were determined, indicating depolymerization of alginates: average molecular weight decreased significantly during the extraction, falling by a factor of 5 between 1 and 4 h of extraction. These results suggested that reducing extraction time could enable preserving the rheological...

Detoxification of Pacific oyster Crassostrea gigas fed on diets of Skeletonema costatum with and without silt, following PSP contamination by Alexandrium minutum ArchiMer
Gueguen, Marielle; Bardouil, Michele; Baron, Regis; Lassus, Patrick; Truquet, Philippe; Massardier, Julie; Amzil, Zouher.
Contamination of shellfish by paralytic shellfish poisoning ( PSP) toxins poses an economic threat to shellfish farmers. As contaminated shellfish cannot be harvested for long periods of time, it would be very useful to develop processes to optimise and shorten their detoxification. In this study, Pacific oysters Crassostrea gigas were first experimentally contaminated over a period of 13 days with a continuous flow of toxic Alexandrium minutum cultures at concentrations ranging from 150 to 200 cell ml(-1) ( toxin content after 13 days of contamination 438 mu g STX equiv. 100 g(-1) wet weight). Then, two different detoxification treatments were tested and showed detoxification rates greater than those observed in coastal environments. The first treatment...

Dynamical modelling of a reactive extrusion process: Focus on residence time distribution in a fully intermeshing co-rotating twin-screw extruder and application to an alginate extraction process ArchiMer
Baron, Regis; Vauchel, Peggy; Kaas, Raymond; Arhaliass, Abdellah; Legrand, Jack.
The context of this study is the modelling of reactive extrusion process based on an alginate extraction protocol. Residence Time Distribution (RTD) is one important part to predict the kinetics of reactive compounds. A simple model is proposed to predict RTD in fully intermeshing co-rotating twin-screw extruders without reaction. This model, which can be easily extended to reactive case in a future work, is based on the extension of an axial dispersion model, including control parameters (screw speed and flow rate) and geometrical parameters (screw profile and die design). Simulations were performed for various operating and geometrical conditions so as to illustrate possibilities offered by the proposed model. Validation was conducted for two different...

Effect of smoking processes on the contents of 10 major phenolic compounds in smoked fillets of herring (Cuplea harengus) ArchiMer
Serot, Thierry; Baron, Regis; Knockaert, Camille; Vallet, Jean-luc.
This study evaluated the effects of five smoking processes on the contents of 10 major phenolic compounds in fillets of herring (Cuplea harengus), namely two traditional processes, involving smoke production by smouldering or friction, one process using liquid smoke atomisation, and two electrostatic smoking processes, also involving smouldering or friction. The effects of several parameters, such as smoking time, smokehouse temperature, electrode voltage, and fish fillet temperature before smoking were assessed for each process. The results indicate that the content of phenolic compounds are strongly affected by the process applied. The percentages of phenolic compounds in fish flesh remained constant and discriminant for a given process, regardless of...

Effects of the smoking process on odour characteristics of smoked herring (Clupea harengus) and relationships with phenolic compound content ArchiMer
Cardinal, Mireille; Cornet, Josiane; Serot, Thierry; Baron, Regis.
The relationship between smoking parameters and odour characteristics, evaluated by a trained sensory panel.. were studied on smoked herring. In addition, a possible correlation between the content of 10 phenolic compounds and sensory perceptions was investigated. Five smoking techniques were applied, combining smoke production conditions, performed by pyrolysis of beech wood sawdust or by friction of beech wood log, with smoke deposition, either in a controlled kiln (traditional smoking) or by an electrostatic process. In the fifth smoking technique, a purified condensate of beech smokes was vaporised on fish fillets in the smokehouse. The time of smoking was 3 It for traditional smoking and the liquid smoke atomisation process and 12 min for the...

Elements for optimizing a one-step enzymatic bio-refinery process of shrimp cuticles: Focus on enzymatic proteolysis screening ArchiMer
Baron, Regis; Socol, Marius; Kaas, Raymond; Arhaliass, A.; Rodriguez Del Pino, J.; Le Roux, Karine; Donnay-moreno, Claire; Berge, Jean-pascal.
This article complements an earlier work published in 2015 Baron et al. (2015) that showed the interest of a shrimp shells bio-refining process. We compare here the effect of eleven commercial proteases at pH 3.5 or 4.0 on a residual amount of shrimp shells proteins after 6 hours at 50 °C. The two pH are obtained when respectively 40 and 25 mmol of formic acid are added to 5 g of mild dried shell. Deproteinisation yield above 95% are obtained. Residual amino acids profile in the solid phase was identical for the eleven proteases except for pepsin which was similar to the raw material profile. A significant relative increase in the proportion of Glycine is observed for the ten other cases. Likewise, shapes of size exclusion chromatograms of the dissolved...

Etude numérique de la répartition de fumée dans une enceinte de fumage industrielle. Rapport final ArchiMer
Havet, Michel; Baron, Regis; Le Niliot, Anne.

First insight on interactions between bacteria and the marine diatom Haslea ostrearia : Algal growth and metabolomic fingerprinting ArchiMer
Lepinay, Alexandra; Turpin, Vincent; Mondeguer, Florence; Grandet-marchant, Quentin; Capiaux, Herve; Baron, Regis; Lebeau, Thierry.
A first insight on the interactions between bacteria isolated from monospecific cultures of the marine diatom Haslea ostrearia and this microalgae is described in this work. From 28 bacterial isolates, the maximal growth rate and the maximal cell concentration were on average 58% and 27% higher (max. 67% for isolate B22). Isolates B5, B10 (both Saccharospirillum sp.) and B22 (Rhodobacter sp.) were the most efficient. The geographical origin of H. ostrearia from which bacteria were isolated had no impact on the algal growth performances, and a given bacterial isolate led to the same algal growth irrespective of the geographical origin of H. ostrearia. To explain the results obtained, a comprehensive integrative strategy was conducted that analyzed the...

Gut passage times in two bivalve molluscs fed toxic microalgae: Alexandrium minutum, A-catenella and Pseudo-nitzschia calliantha ArchiMer
Gueguen, Marielle; Lassus, Patrick; Laabir, Mohamed; Bardouil, Michele; Baron, Regis; Sechet, Veronique; Truquet, Philippe; Amzil, Zouher; Barille, Laurent.
The occurrence of new phytoplankton species in a coastal area may be explained by the import of shellfish containing whole live algal cells in their digestive tracts. Indeed, shellfish containing toxic algal cells can induce both primary contaminations in safe areas ( initially free from toxic microalgae), and secondary contaminations of other shellfish growing in the same area via the faeces of the imported animals. To mitigate this problem, shellfish need to be placed in a separate holding tank and their intestinal content purged. For a deeper understanding of the risks associated with transferring contaminated shellfish, oysters ( Crassostrea gigas) and mussels ( Mytilus edulis) were purposely fed either Alexandrium minutum or A. catenella (...

Improving detoxification Efficiency of PSP-contaminated oysters (Crassostrea gigas Thunberg) ArchiMer
Lassus, Patrick; Bardouil, Michele; Baron, Regis; Berard, Jean-baptiste; Masselin, Pierre; Truquet, Philippe; Pitrat, Jp.
Shellfish production, and production of marine bivalve molluscs in particular, is the main form of marine aquaculture in the European Union, with a production estimated at 1,200,000 tons in 1999 (Food and Agricultural Organization). Moreover, aquaculture production is rapidly expanding throughout Europe and is becoming a major economic resource in developing areas such as Northern Greece, Ireland, and northwestern Spain. These shellfish, mainly mussels, oysters, and clams, are produced in a natural environment. Due to variations in their environment, these otherwise edible species may however become unsafe for human consumption. Potential problems are amplified due to the nature of shellfish themselves: as they are filter feeders, they consume large...

The purpose of this study was to assess paralytic phycotoxin uptake in diploid and triploid oysters at two stages of their sexual cycle corresponding to their status in early summer (June) and winter (November). Samples of diploid and triploid oysters were exposed to a toxic culture of Alexandrium minutum for 4 days in each season. No significant differences in filtration or clearance rates were observed during either November or June experiments. When diploid oysters were at resting stage (November), toxin uptake showed no significant difference between the ploidy classes. In contrast, when the diploid oysters were at the peak of their sexual maturation (June), the triploid oysters were seen to accumulate almost double the amount of paralytic toxins as...
